As long as there is strong contrast between the dots and the background - dark on light works best - it will scan reliably. Very light dots or low contrast can fail, so test before printing.
Use PNG for general digital use and printing - it always exports with a transparent background. Use SVG when you need a scalable vector that stays sharp at any size, and JPG when a format without transparency is required.
